Production of siRNA-Loaded Lipid Nanoparticles using a Microfluidic Device

The development of functional lipid nanoparticles (LNPs) is one the major challenges in field drug delivery systems (DDS). Recently, LNP-based RNA systems, namely, RNA-loaded LNPs have attracted attention for therapy. In particular, mRNA-loaded LNP vaccines were approved to prevent COVID-19, thereby leading paradigm shift toward next-generation nanomedicines. For nanomedicines, size a significant factor controlling biodistribution and performance. Therefore, precise control technique indispensable production process. Here, we report protocol controlled using microfluidic device, named iLiNP. siRNA loaded are also produced iLiNP device evaluated by vitro experiment. Representative results shown size, including siRNA-loaded LNPs, Z-potential, encapsulation efficiency, cytotoxicity, target gene silencing activity.
